The Possible Protective Effect of L-Arginine Against Aspartame-Induced Toxicity on Adrenal Cortex of Adult Male Albino Rats: Histological and Immunohistochemical Study

Abstract
Aspartame is one of the frequently utilized artificial sweeteners. It has been linked to numerous degenerative diseases and oxidative damage, whereas L-arginine, a semi-essential A.A., has antioxidant properties via the production of Nitric oxide, which is involved in the modulation of tissue perfusion and cell proliferation.

The aim of this work was to elucidate the probable protective effect of L-arginine against adrenal cortex toxicity induced by Aspartame.

Material and Methods: Fifty adult male albino rats were divided into the 5 groups (each comprised 10 rats): Control group I: were held without medication for 4 weeks. Group II: received L-arginine, at a dose of 0.3mg/gm/day for 4 weeks. Group III: received Aspartame, at daily dose 250 mg/Kg for 4 weeks. Group IV: received Aspartame and L-Arginine as previously for 4 weeks. Group V: received Aspartame, at daily dose 250 mg/Kg for 4 weeks and then were held without medication for another 4 weeks then sacrificed. Just before sacrifice, blood samples were drawn for biochemical assessments. Adrenal cortex sections were subjected to histological, immunohistochemical, and morphometric studies. Aspartame resulted in significant deterioration of histological architecture of adrenal cortex and L-Arginine mostly reversed these changes. Aspartame led to significant increase in Caspase-3 immunohistochemical area as compared to control group. On the other hand, Group IV revealed marked histologic and functional improvement. The recovery group (V) revealed incomplete improvement.

Conclusion: L-arginine can prevent the Aspartame induced oxidative stress in adrenal cortex in comparison with the untreated group may be through antioxidant effect.
